BACKGROUND: Considerable attention has been paid to plant‐growth‐promoting rhizobacteria (PGPR), especially the fluorescent group of Pseudomonas species, as the best alternatives to chemicals for facilitating ecofriendly biological control of soil‐ and seedborne microorganisms. On the basis of their novel plant‐growth‐promoting attributes, two rhizobacteria Pseudomonas aeruginosa VP1 and VP2 selected out of over 63 isolates from the rhizosphere of chilli (Capsicum annuum) were identified as potential candidates for biocontrol of the root‐knot nematode Meloidogyne incognita on chilli. RESULTS: The nematicidal activity of both strains was evaluated in vitro and in vivo for their efficacy against M. incognita. P. aeruginosa VP2 exhibited strong nematicidal activity in comparison with VP1, based on the in vitro killing of the second‐stage juveniles (J2) of M. incognita. Seed bacterisation with both strains VP1 and VP2 was able to manage root‐knot M. incognita on chilli (C. annuum) in a pot trial study. Increase in root and shoot length and in fresh and dry weight of root and shoot and reduction in the root‐knot index over the control were attained. In overall performance, VP2 was 29.5% more effective than VP1, and about 30% more effective than the control (non‐bacterised). CONCLUSION: The application of P. aeruginosa VP1 and P. aeruginosa VP2 controls the development of M. incognita in C. annuum, and hence they are recommended as efficient plant growth promotors and biocontrolling agents for raising healthy crop of C. annuum that can promote the growth of plants and reduce the nematode (M. incognita) population. Bioconverted eicosapentaenoic acid (bEPA), obtained from Pseudomonas aeruginosa PR3, was assessed for its in vitro and in vivo anti-fungal potential. Mycelial growth inhibition of the tested plant pathogens (Rhizoctonia solani, Botrytis cinerea, Fusarium oxysporum, Fusarium solani, Phyptophthora capsici, Sclerotinia sclerotiorum and Colletotrichum capsici) was measured in vitro. bEPA at the concentration of 5 μl/ml inhibited 52–60% fungal mycelial growth for all of the plant pathogens in vitro except S. sclerotiorum. Minimum inhibitory concentrations (MICs) of bEPA were found in the range of 250–500 μg/ml. Also, bEPA had a detrimental effect on spore germination for all the tested plant pathogens. Three plant pathogenic fungi (F. oxysporum, P. capsici and C. capsici) were subjected to an in vivo anti-fungal screening. bEPA at the initial concentration of 3000 μg/ml had a 100% anti-fungal effect against all of the tested plant pathogens. Concentrations of bEPA corresponding to 1500, 500 and 300 μg/ml were applied to the plants and revealed promising anti-fungal effects, supporting bEPA as a potential anti-fungal agent.  相似文献

PGPR strain Pseudomonas fluorescens PS1 was evaluated to formulate carrier based bioformulations. The viability of P. fluorescens PS1 was monitored at different time intervals during the period of storage at room temperature in different carriers such as soil, charcoal, sawdust and sawdust-soil. Sawdust-soil was found to be the most efficient carrier material for P. fluorescens PS1 followed by other carriers. After 1 year of storage, P. fluorescens PS1 was re-isolated and assayed for its antifungal activity against Sclerotinia sclerotiorum a phytopathogenic fungus causing stem blight in Indian mustard, Brassica campestris. Results of scanning electron microscopy exhibited that P. fluorescens PS1 caused morphological alteration in mycelia of S. sclerotiorum as evident by hyphal perforation, and fragmented lysis. Seed bacterization of B. campestris with P. fluorescens PS1 induced enhanced seed germination, increased overall plant growth as well as reduced stem blight in mustard with improved yield. These findings demonstrate that P. fluorescens PS1 has significant potential to raise disease-free crops due to the presence of a wide array of PGP characteristics.  相似文献

This study is focused on investigating the feasibility of using silver(I) ions loaded poly(acrylonitrile)-grafted silk fibers as antibacterial dressing material. The optimum grafting conditions for ceric ammonium nitrate induced graft-copolymerization of acrylonitrile onto silk fibers were found to include initiator concentration of 35 mM, catalyst HNO3 concentration of 0.40 M and initiation time of 10 min. The poly(acrylonitrile)-grafted silk fibers were loaded with silver(I) ions by equilibration method. The resulting fibers were investigated for their biocidal action against E. coli, by using zone inhibition and colonies counting method.  相似文献

Powdery mildew caused by the ascomycete Phyllactinia guttata (syn. P. corylea) is a major foliar disease worldwide of the unique mulberry (Morus spp.) for silkworm feed. Genetic resistance to powdery mildew, the most sustainable and economical strategy for disease control, is still elusive for tropical mulberry. About 147 germplasm sources, representing 18 countries of origin, were screened for resistance to P. guttata in six seasonal fields and greenhouse trials after exposure to natural and artificial inoculum, respectively. In the field, the level of plant responsiveness to disease was assessed from 30 to 62 days after pruning in each season as variations in the disease severity index (DSI), disease incidence (DI%) and area under the disease progress curve (AUDPC). These measures differed significantly among the germplasm. Of 147 germplasm sources, ~6.8% had useful resistance (two high and nine moderately resistant) to the powdery mildew pathogen on the basis of DSI. The AUDPC values were 13.5-fold higher in the most susceptible accession—(Philippines) than the least responsive (Vietnam-2). The results of DSI were strongly correlated with the obtained DI values (r = 0.92; P < 0.01) and AUDPC (r = 0.89; P < 0.01). Moreover, field screening results were highly correlated (R 2 = 0.839) with values from the greenhouse evaluation using artificial inoculum. However, the DSI values in field and greenhouse screenings for three sources (Non-nayapati, Nao-khurkul and Tista Valley) varied significantly. A relatively low disease reaction of 09 resources (Vietnam-2, Ankara and 07 others) using different assessment scales after natural and artificial inoculation prove, for the first time, that they have potential in breeding for resistance in tropical mulberry to powdery mildew.  相似文献

Pentachlorophenol (PCP) adsorption and desorption equilibrium was studied with two Menfro silt loam soils — upper horizon and lower horizon. For the adsorption studies the variables were: temperature (10 and 30 °C) and the amount of organic matter. The variables for the desorption studies were: temperature (10 and 30 °C), pH and the presence of an anionic and a cationic surfactant. The results from these studies confirmed the importance of soil organic matter for adsorption of PCP on the soils. The adsorption data at different temperatures indicated the physical nature of the adsorption process. The desorption data produced non-singularity and some PCP was irreversibly adsorbed onto the soil despite repeated washings. Increased pH increased the desorption of PCP from the soil. The anionic surfactant, sodium dodecylbenzene sulfonate (SDS) was able to desorb significant amounts of PCP from the soil at doses equal to critical micelle concentration (CMC). But, the nonionic surfactant, surfactant, Triton X-405 required a much higher dose (twice the CMC) to cause a significant desorption of PCP from the soil.  相似文献

Seedlings from six crosses between susceptible × resistant and two between susceptible parents were screened against P3 isolate of Phytophthora drechsleri f. sp. cajani in the glasshouses. Disease reaction was scored on a rating scale of 1–5. Resistance was found to be controlled by one dominant gene and some minor genes and is affected by ontogeny. The resistance gene appeared to be different from the one reported earlier (Pd1) and is designated here as Pd3. In this study cyclodextrin (CD) has been grafted onto cellulose back-bone of cotton fabric using citric acid as crosslinker. The CD-grafted fabric was characterized by SEM and FTIR analysis. The percent grafting of CD on fabric has been found to increase with concentration of CD, citric acid, and sodium dihydrogen phosphate. The CD-grafted fabric has been loaded with silver(I) ions for the purpose of obtaining a slow release device. The Ag(I) loaded grafted fabric showed fair antibacterial properties against E. Coli. The release of Ag(I) ions from the CD-grafted fabric was observed for a period of seven days.  相似文献

In order to study the effect of feeding graded levels of green vegetables on egg production performance and egg yolk antioxidant status, 27 female golden pheasants (GP) were randomly distributed into three groups of nine each in an experiment based on completely randomized design. The diets of the birds in groups T1, T2 and T3 contained 1.4%, 2.7% and 5.0% of green vegetables respectively. Feeding of experimental diets started on 12th February (day 1) and was continued till 30 June 2012. Average number of eggs laid and egg mass produced by the hens in group T3 was higher (p < 0.004) than those of T1 and T2. Hen day egg production was lowest (p < 0.001) in group T1. Roche yolk colour score was highest (p < 0.01) in T3 followed by T2 and was lowest in T1. Other external and internal egg quality parameters were similar among the groups. Total antioxidant capacity (TAC) of egg yolk was higher (p < 0.04) in group T3 as compared to other two groups. Egg yolk concentration of total carotenoids, β‐carotene, total sulfhydryl, protein‐bound sulfhydryl and non‐protein‐bound sulfhydryl was higher (p < 0.05), whereas concentration of malondialdehyde was lower (p < 0.024) in group T3 as compared to other two groups. Excreta concentration of corticosterone was highest (p < 0.012) in T1, followed by T2, and was lowest in T3. Egg yolk concentration of TAC and TSH was positively and that of malondialdehyde was negatively correlated with carotenoids intake. Excreta concentration of corticosterone was negatively correlated with carotenoids intake. It was concluded that incorporation of green vegetables in the diet at 5% on dry matter basis would improve egg production, egg yolk antioxidant status and the ability of the captive GP to combat stress.  相似文献
